Abstract

Cost-effective screening for oral bioavailability using the isolated perfused rat intestine

Recently, commercial and ethical pressures have increased the demand for higher throughput and lower cost methods for screening candidate compounds for bioavailability and toxicity, and which also give maximum information for minimum animal usage. Isolated perfused organs have a number of distinct advantages over in-vitro techniques such as the CaCo-2 cell and hepatocyte in that the component cells are exposed to the drug at concentrations mimicking those in the intact animal. In addition the cellular makeup of the organs remains intact and retains full metabolic capability.

The isolated perfused rat intestine model enables us to rank order compounds for bioavailability using a cassette dose of typically 5 compounds. The preparation can also be used to investigate the specific problem of apparent low oral bioavailability enabling us to distinguish between poor absorption and metabolism by the liver and/or gut. This rapid cost-effective methodology can be used for screening of candidate compounds for bioavailability with minimal animal usage.
