The isolated perfused bovine uterus as a model for mucous membrane irritancy and inflammation

Bovine uteri of slaughtered healthy cows are perfused with heparinized and oxygenated tyrode solution (flux 18 ml/min) or blood. The viability of the organs is demonstrated by nearly unchanged lactat production, lactate dehydrogenase activity and glucose consumption. In first experiments, irritating effects of antiseptics which are frequently used topically in endometritis treatment of cows were studied. It was demonstrated that the viability of the mucosa measured with a modified methyl tetrazolium assay (MTT) reflects the concentration dependent irritating potency of the test compounds. Additionally, the isolated perfused bovine uterus is used as a model of inflammation. Hemoperfused bovine uteri are inflamed topically by arachidonic acid, carrageenin and lipopolysaccharide administration. The measurement of mediators of inflammation (eicosanoids, cytokines) demonstrates the inflammatory reaction.
